Parling is named among the replacements for the game, with his Leicester Tigers teammates Dan Cole, Tom Croft and Ben Youngs given starting spots.
The uncapped Parling was picked for England’s tour of Australia in 2010, where he played in a tour game against Australian Barbarians.
The 28-year-old lock is one of eight uncapped players in the England matchday 22, three of whom start the game. Saracens Brad Barritt and Owen Farrell form a new centre pairing, while Northampton Saints’ Phil Dowson makes his debut at No 8.
Parling is joined on the bench by four more uncapped players in Rob Webber (London Wasps), Ben Morgan (Scarlets), Lee Dickson (Northampton Saints) and Jordan Turner-Hall (Harlequins).
Head Coach Stuart Lancaster said: “We have been impressed with how the whole group has come together in the last 10 days and we have done a lot of work on and off the field.
“Now is the time to turn all that into a performance against Scotland that we can all be proud of. It is a massive challenge – Murrayfield, the first game of the Six Nations – but one we are relishing.”
The England match will be shown live on the screens around Welford Road, including a big screen in the Final Whistle, on Saturday (5pm kick-off) after the Tigers versus Newcastle Falcons game in the LV= Cup.
