Prop Neale Farthing played as a second-half replacement in last Saturday’s 57-13 victory over Scotland and is now joined in the squad by club colleagues Will Owen, George Tressider and Tom Hudson.
Hudson starts at full-back, with flanker Owen also in the starting line-up, while Farthing and scrum-half Tressider are among the replacements.
The quartet all played in Leicester Tigers Academy’s successful season in the Premiership Academy Under-18 League.
England’s January 1st side enjoyed a good run out against Scotland last week, scoring nine-tries in their win in Darlington, and nine members of that matchday squad are in line to face Wales.
Head coach Fletcher said: “Playing in Wales is always a challenge but it’s one we’re looking forward to. This is our September 1st team so we’ve been able to bring a few more experienced players into the matchday squad and we’re looking forward to seeing how they get on.”
