Centre Simpson-Daniel, who scored twice in a four-minute spell against Bath last Saturday, is ruled out by a side injury.
The other change to the team that beat Bath 36-27 sees a return for Luke Narraway, who comes in for Apo Satala who is with Fiji at the World Cup Sevens.
“To lose a player of Simpson-Daniel’s quality for a game of this magnitude is significant because he is such an attacking threat,” said head coach Dean Ryan.
“Bath played their part last Saturday but for us it was confirmation that things we have been working on are starting to take shape. There were fundamentals in our game that turned up last weekend and enabled us to execute our game.
“Those are the levels that are demanded again tomorrow against a very good side.”
