Tigers won the toss and had choice of colours and dressing rooms for the final against Ospreys, who will wear their black kit.
Officials are expecting a crowd of more than 60,000 for Saturday's match after advanced sales passed 58,000 this week - well up on last year's final attendance of 42,538.
Tickets have now been returned from Tigers, but they are available on the day at Twickenham.
RFU business operations director Paul Vaughan said: “The expected crowd shows how important cup competitions are to the rugby supporter. With the Tigers aiming to retain the trophy and the Ospreys smarting after their Heineken Cup exit, it’s going to be a fantastic occasion.”
Saturday’s action begins with the EDF Energy Intermediate Cup between Birkenhead Park and Chester at 10am, followed by Exeter Chiefs against Northampton Saints in the EDF Energy National Trophy at noon and then Tigers against Ospreys at 2.30pm.
In the event of the scores being level after 80 minutes, extra time will be played. If the scores are level after extra-time then the club with the most tries shall be deemed winners. If the number of tries is equal, then the winner will be determined by a place-kicking competition.
Tigers Travel Club coaches for more than 1,000 supporters will be leaving Welford Road at 9am on Saturday. Coach numbers are printed on your travel tickets.
Supporters should be aware that, due to the number of coaches involved, there is no parking available on the club premises behind the Next Stand. Please use public car parks.
The Tigers Club Shop at Welford Road will be open from 7.45-8.45am and a mobile shop unit will be open in the West Car Park at Twickenham from 10am.
Ref!Link radios are in operation at Twickenham, but work on a different frequency to the sets used at Welford Road. If you present your blue Ref!Link radio to the kiosks at Twickenham, you can make a 20% saving and buy a Twickenham set for £4 instead of the standard price of £5.
