Tigers play Saracens in the showpiece event on Saturday in front of a sell-out 81,000 crowd.
At half-time 10 pupils from Parks Primary School in Leicester will play in a tag rugby match on the famous Twickenham turf as part of Tigers’ Something To Chew On programme.
Mary Yates, teaching assistant at Parks Primary School, said: “The children can’t believe it.
“It is an amazing opportunity. They do realise it’s a once in a lifetime opportunity for the children who are going. They are all really excited.”
Something To Chew On is delivered by Tigers in partnership with Premier Rugby and the Food Standards Agency.
The programme sees Tigers community coaches deliver tag rugby sessions and classroom-based lessons on healthy eating and nutrition.
Young players from Burton on Trent and Nottingham Boots Corsairs rugby clubs will also be at Twickenham on Saturday for a Parade of Champions after winning the Tigers Land Rover Premiership Cup festival in April, held at Oakham RFC.
Burton won the under-11s tournament while Nottingham Boots Corsairs were the under-12s champions.
Both teams will join a Parade of Champions with the winning teams from around the 12 Guinness Premiership clubs.
Burton have 70 people travelling down for the occasion and Martin 'Rita' Hulbert, chairman of Burton mini and juniors section, said: "I think the adults are more excited than the lads, if that can be possible. We are so excited and proud of our children.
"The fact that the Tigers are playing is the icing on the top for them."
The Land Rover Premiership Cup is a national grassroots tournament officiated by Premier Rugby and run in conjunction with the 12 Guinness Premiership clubs.
