Harry Ellis, George Chuter, Louis Deacon and Martin Corry all start, while Julian White and Lewis Moody claim a place on the bench for the Calcutta Cup clash.
Tigers have more players in both the starting XV and the 22 than any other club, with London Wasps next in line with a total of four players making the match-day squad.
Corry , along with Mike Tindall and Phil Vickey, is in line to win his 50th international cap, with Ellis making his first England appearance since last year's 6 Nations tournament.
Tindall also returns to the team for his first international since the 2006 competition and Vickery leads the team for the first time in a full test at Twickenham.
Saracens inside centre Andy Farrell is set for his England debut in a side which also sees the return from test retirement of Jason Robinson and a first international start for Jonny Wilkinson (Newcastle Falcons) since the RWC 2003 final.
Ashton said: "I'm delighted to announce a strong England 22 for what will be a very challenging match against Scotland on Saturday. The starting XV includes a debut for Andy Farrell. It is well documented that I have been an admirer of Andy 's career for a number of years and I'm looking forward to seeing him play rugby union for his country.
"I'm also pleased that Jason Robinson has decided to return from international retirement and that Jonny Wilkinson has returned to fitness after his recent injury and that both are available for the Scotland game."
