Tigers go to France to face Clermont in the first leg on Sunday, April 9 before the return on home turf on Saturday, April 16 (5.30pm).
The winners will be determined by aggregate scores over the two games, with the winners paired against Leinster or Connacht in the next round.
Clermont and Tigers have been familiar foes in Europe, with meetings in the pool stages and in knockouts, though not since a quarter-final in France during 2014.
Benjamin Kayser, Seremaia Bai, Kini Murimurivalu, Pat Howard and Richard Cockerill are among the players who have represented both clubs, while Argentina international Tomas Lavanini swapped Welford Road for the Stade Marcel-Michelin last summer.
All season ticket holders have had this fixture added to their season tickets are no cost following cancellation of the game against Bordeaux-Bègles earlier this season.
