Stratford-upon-Avon will take on Wymondham and Burton will face Lutterworth at the home of Leicester Tigers before the Guinness Premiership game against Worcester Warriors.
Scott Clarke, Tigers community manager, said: "The semi-finals look set to be a great occasion and I am sure the players will be giving their all in training this week.
"We are encouraging Tigers supporters to get to Welford Road early to cheer on the teams and we wish everyone involved the best of luck."
Lutterworth are going in search of their fourth Prima Tiger Cup title, while the other three finalists have yet to win the prestigious tournament.
Wymondham's preparations for their game against Stratford will see them stay in Leicester on Friday night before training on Nelson Mandela Park, opposite Welford Road, where some illustrious former Tigers have played and trained in the past.
Dominic Findlay, head coach of Wymondham under-10s, said: "We came up on the Friday before the quarter-final game on the Saturday and we are doing the same this time. We hope it works again but you just don't know."
The Prima Tiger Cup, which is supported by Loughborough-based Prima Solutions, is now in its 15th season and is open to under-10s teams from across the country.
More than 50 sides entered this year's competition, which began with local festivals at Stoke RFC in Staffordshire, Kenilworth RFC in Warwickshire, Swaffham RFC in Norfolk, Nottingham Boots Corsairs RFC and at Market Harborough RFC in Leicestershire.
The final will be held at Welford Road before Tigers' Guinness Premiership game with Gloucester on Saturday, March 7.
